Summary:Indonesia is known as megacenter of biodiversity. Thousands of species inhabit in this country region. One of the endemic species is the Tarsier. This world's smallest primate is one of the wealth of this nation but the existence and survival of Tarsius sp. is threatened. The era of globalization and modernization demanded all the most detailed information and in-depth study including molecular and DNA analysis. The main objective of this research is to analyze the genetic diversity Tarsius bancanus borneanus, Tarsius spectrum, and Tarsius dianae based on sequences of NADH dehydrogenase sub-unit 1 (ND1) encoding gene. Amplification of ND1 gene by PCR technique used the result of DNA isolation, primer ND1 Forward and primer ND1 reverse. The PCR reaction produced 957 bp DNA. The result of ND1 gene sequences then multiple aligned with other primates based on source data from Genbank. Analized result using MEGA version 5.2.2 showed there were nucleotides and amino acid variation between Tarsius bancanus borneanus, Tarsius spectrum, and Tarsius dianae. Genetic distances based on nucleotide sequence of ND1 genes analyzed using Kimura 2-parameter is 13 %. Tarsius sp. included in suborder Strepsirrhini based on construction of phylogenetic tress used Neighbor-Joining method based on nucleotide sequence and amino acid sequence of ND1 gene with bootstrap values 1000 times.
