PENGARUH PEMBERIAN TAMOXIFEN PADA HATI TIKUS SPRAGUE DAWLEY PASCA OVARIEKTOMI

This research was aimed to study the effect of tamoxifen on body weight, liver weight to body weight ratio, macroscopic and histopathological figure of Sprague Dawley rat liver two months after ovariectomy. A total of 8 female rats (Sprague Dawley) were divided into 2 group of 4, group A was ovariectomized rats without administration of tamoxifen and group B was ovariectomized rat with administration of tamoxifen 10 mg / kg body weight. Tamoxifen was given orally for 2 months. Rats were then necropsied to take the liver. Liver were taken and weighed first, then put it in 10 % formalin solution for histopathological examination using Hematoxyline Eosin staining. The results showed that the body weight of ovariectomized rat without tamoxifen administration (203.5±12.1g) was significantly higher than ovariectomized rats with tamoxifen administration (182.5±6.5g) (P < 0.05). Ratio of liver weight to body weight of ovariectomized rats with tamoxifen administration (0,035 ± 0,0021g) was significantly higher than ovariectomized rats without tamoxifen administration (0,032±0,00096g) (P<0.05). Macroscopic figure of the liver of ovariectomized rats with tamoxifen administration was not shown any changes, however, there was fatty degeneration and necrosis histopatologically. Based on the results, it can be concluded that the administration of tamoxifen (10 mg / kg body weight) might affect body weight, liver weight to body weight ratio and histopathologic figure of liver 2 months after ovariectomy, however macroscopic figure of the liver was not affected.
