الملخص: | Chitosan is a natural polymer and it is biodegradable polysaccharide, nontoxic,
unfold tight junction, mucoadhesive and increase drug absorbtion. Long
chains chitosan has properties that are less profitable, because of its poor solubility
in water and high viscosity. Thus, in this research, chitosan chain scission with 150
kGy radiation dose of gamma rays. This study aims to produce GVT-0
nanoparticles using irradiated chitosan with ionic gelation method and
tripolyphosphate as a crosslinker.
Characterization of Gamma-rays irradiated chitosan are organoleptic,
determination of its viscosity, and degree of deacetylation, meanwhile the
characterization of GVT-0 nanoparticles include particle size, particle size
distribution, morphology of nanoparticles, and entrapment efficiency.
The results showed that the irradiated chitosan has more brownish color flake,
lower viscosity (2,26 ± 0,12 cP) and higher degree of deacetylation (65,230%)
compared with non-irradiated chitosan (viscosity: 86,78 ± 4,008 cP and a degree of
deacetylation: 65,230%). Irradiated chitosan can produce GVT-0 nanoparticles that
has better characteristics than non-irradiated chitosan. GVT-0 nanoparticles
produced by irradiated chitosan has smaller particle size (48,967 ± 4,126 nm), more
uniform particle size distribution, spherical shape, but has lower entrapment
efficiency (93,048% ± 0,033%) at the formula C2 with concentration of GVT-0:
chitosan: TPP = 0,02% - 0,04% - 0,003%, compared with GVT-0 nanoparticles
produced by non-irradiated chitosan. |
