Summary:Iguana iguana is one member of a family of Iguanas (Iguanidae) and classified as herbivorous reptiles. Easy handling of Iguana iguana makes a lot of people began to choose this animal as a pet. One of the problems often encountered in the maintenance is error feeding. The knowledge of feed is in close association with the digestive system. Thus, knowledge of the anatomical structure of the digestive system is essential for proper feeding patterns. This study aims to determine the histological structure of the digestive tract, particularly the stomach of Iguana iguana. The study use three iguanas. Iguana iguana were put to sleep using chloroform and then dissected. Stomach tissue were observed macroscopically and fixed in Bouin's solution. After 24 hours in Bouin's solution, the sample was transferred to 70% alcohol. Furthermore, organ samples processed by the paraffin method, cut 5μm, and then stained by HE and AB-PAS method. Results were analyzed descriptively. Stomach of Iguana iguana were divided into three parts, namely cardiac, fundic and pyloric. Lamina epitelialis mucosae on the entire surface of stomach is a columnar epithelium simplex. Cardiac glands are large and rare, the fundic gland are small-size and compact, while pyloric glands are medium-size and compact. Cardiac and pyloric gland consist of cuboidal-columnar mucus-secreting cells, whereas fundic gland dominated by round-shaped cells with round nuclei in the middle. This cell suggested as oxyntico-peptic cell. The muscularis layer of cardiac and pyloric area consist of circular smooth muscle, but in the pyloric divided into internal-circular muscularis layer and external-longitudinal muscularis layer. The AB-PAS staining showed a negative reaction to AB, but positive to PAS on the surface of epithelial cells, gastric pit and gland cells.
