Summary:Doxorubicin as one of the most commonly used cancer chemotherapy agent is associated with many adverse effects such as immunosuppression and cardiotoxicity. Polysaccharide from noni fuit (Morinda citrifolia L.) was known to possess anti-tumour and immunomodulatory effect. Combination of doxorubicin with noni fruit polysaccharide may improve immune system integrity while retaining their anti-tumour and tumour-suppressing effect. This study investigates cytotoxicity effect, selectivity, and apoptosis-inducing effect of the combination against HeLa cell line. Combination of doxorubicin with 4 different concentrations of polysaccharide-rich fraction from noni fruit was tested in vitro with MTT-reduction assay against HeLa and Vero cell line to determine the enhancement of cytotoxic effect from doxorubicin by noni fruit polysaccharide and their selectivity. Apoptotic assay using ethidium bromide-acridine orange staining were performed to find out apoptosis-inducing effect of the combination. the composition of the fraction used were also analyzed qualitatively. The IC50 score of doxorubicin against HeLa cell line was obtained at 4 μM from the assay. Combination of polysaccharide-rich fraction at 100 μg/mL and doxorubicin at 2 μM could induce the highest cytotoxic effect that differs significantly from single doxorubicin treatment at the same concentration. The combination given proved to be able to induce apoptosis on HeLa cell line and apparently more cytotoxic against HeLa than Vero cell line.
