Summary:Chitosan has been known as an antibacterial agent which is biodegradable, biocompatible, but insoluble in neutral solvents. Modification into N,O-carboxymethyl chitosan (N,O-CMC) was done in order to increase solubility in neutral solvent without changing its bioactivity so application as an antibacterial agent against fish disease bacteria is allowed. Three methods were employed to produce N,O-CMC. Characterization was done in order to know the important characteristics of chitosan and its derivate, i.e. yield, solubility, chemical bond and degree of deacetylation. The result in characterization showed increasing in solubility and degree of deacetylation. The three products were tested against fish pathogens Aeromonas hydrophila, Streptococcus sp. and Vibrio spp. Two types of N,O-CMC products have antibacterial activity against Streptococcus sp., and Vibrio spp. None of N,O-CMC products showed antibacterial activity against Aeromonas hydrophila. The best concentration of Liu�s N,O-CMC and Hayes�s N,O-CMC against bacteria in this experiment is 20 mg/mL.
