The aim of this research was to test the haemolytic effect of Trimeresurus puniceus venom on dog erythrocytes. The materials used in this research were Trimeresurus puniceus venom and dog erythrocyte. The venom was diluted with phosphate buffer saline (PBS) into concentrations of 10%, 1% and 0.1% whereas the concentration of dog erythrocyte used was 100%. The tests were done by mixing 10μL of dog erythrocyte with 10μL of venom of each concentration for 5 minutes. The resulting damage to erythrocytes was counted using a haemocytometer. From the research, the venom with 0.1% concentration caused light changes to the erythrocyte cell wall making it irregular, causing damage to roughly 58% of erythrocytes. The venom with 1% concentration caused crenation and lysis of erythrocytes, with roughly 76% of erythrocytes undergoing lysis. The venom with 10% concentration caused lysis of erythrocytes, with roughly 81% of erythrocytes undergoing lysis. Statistically, 0.1%, 1.0% and 10.0% venom concentration causes significant damage to erythrocytes of the dog. This shows that, the venom contains hemotoxic and cytotoxic properties.
