Summary:Johar or Cassia siamea Lamk. classified as plant which is easily grow in tropical country like Indonesia. Barakol in johar leaves has anxiolytic effect for sleeping disturbance. This condition can be used by traditional medicine business person. The choice of extraction method is an important thing to obtain optimal active compound. This research is carried for knowing extraction method which resulted maximum content of barakol. Johar leaves powder is extracted by macerating for 24 hours and infusing for 30 minutes, until the concentrated extract obtained. Maceration extract (M) is fractioned by n-hexane and infudation extract for 30 minutes, known decoct (D), fractioned by ethanol 96%, so that n-hexane soluble fraction (FLH), n-hexane insoluble fraction (FTLH), ethanol soluble fraction (FLE), and ethanol insoluble fraction (FTLE) are obtained. Relative content of barakol is measured by TLCdensitometry with isolated barakol as reference. Chloroform:ethanol (9:1) is used as mobile phase and silica gel F254 is used as stationary phase. The barakol content is analyzed with one way ANOVA in convidence level of 95%. The difference in extraction method and fractionation effects extracted barakol relative content. Effective extraction method which resulted highest barakol relative content is maceration in ethanol 96% without fractionation.
