Summary:The objective of study was to characterize protease of degrading feather chicken isolate bacteria. Proteolytic bacteria isolated from the soil in the poultry slaughter waste area (B2 isolate) was used to produce enzyme at the temperature 50°C, pH 8 for 4 days. Crude enzyme obtained was presipited with 70% saturated Ammonium Sulphate and dialyzed in phosphate buffer solution. The enzyme activity was characterized. The observation was done at the incubation temperature of 30, 33, 35, 37 and 40°C, medium pH at 6.0, 6.5, 7.0, 7.5 and 8.0, substrate concentration at 0.1, 0.25, 0.5, 0.75, 1.0, 1.25 and 1.5% and also time of incubation at 10, 20, 30, 40, 50 and 60 minutes. Kinetic parameters (Km and V.x values) were analyzed its optimum conditions. Highest protease spesific activity was observed at temperature of 35°C (0.085 U / mg prot), pH 7.0 (0.10 U /mg prot), substrate concentration of 1.25% ( 0.125 U / mg prot) and incubation time of 60 minutes ( 0.148 U /mg prot) (P
