Summary:Abstrak A research was conducted to evaluate the effects of the 2,3,7,8 tetrachlorodibenzo-p-dioxin (TCDD) on the immune response of neutrophils and lymphocytes of rats. Sixteen male Sprague-Dawley rats, 2-2.5 months of age and 170-200 g of body weight were adapted to the research environment for one week. All rats were randomly allotted into 2 groups of 8 rats. The first group was given 1 ml/kg bw/oral/day of PBS pH 7,2 as placebo for 60 days. The second group was treated orally with 2,3,7,8 TCDD 5 mg/kg bw on the first day and followed by 1 mg TCDD/ kg bw/day for 60 days. On the day of 30 and 60,4 rats in each group were randomly selected and blood samples were collected from retroorbital plexus. The neutrophils and lymphocytes were separated by the gradient density methods using Histopaque-1077 for the study of the immune response. The phagocytic activities of neutrophils to Staphylococcus aureus were assayed with acridine orange fluorescence method. The proliferation activities of lymphocytes were evaluated by the stimulation of lymphocytes with Con-A. The data of neutrophil phagocytic and lymphocyte proliferation activities were analysed with the 2x2 factorial anova followed by t-test. The neutrophil peroxidase activities were analysed discriptively by compairing the treated to control animals.
