There are three different kinds of rhizomes as indicated of 'Kunir Putih' in Yogyakarta, those are: Curcuma zeodaria, C. longa and C. mangga . It was known that C. zeodaria and C. longa induced apoptosis on cell lines, but not C. mangga yet. Previous research showed that he potential contents of C. mangga Val are volatile oil and non-oil secundary metabolites. The objective of this research is to find out the in vitro effect of the volatile oil of C. mangga on the cytotoxicity, antiproliferation, and apoptosis of Raji and Myeloma cell lines. Three different samples of C. mangga oils (oils of main rhizome, branch rhizome, and mix of both rhizomes) were used for the research. The in vitro cytotoxic effect was examined by staining the viable treated Raji and Myeloma cells using trypan blue. The proliferation kinetic was determined by counting the cell number in time courses to determine the doubling time of the cells. Apoptotic proscess was examined by observaring microscopally the cell morphology that was stained usingethidium bromide as well as by electrophoresis and immunocytochemistry. The results showed that the mix volatile oil of C. mangga using concentration of 125 µl/ml produced the highest cytotoxic effect on Raji and Myeloma cell lines. The antiproliferation assay showed that C. mangga oil inhibited the growth of Raji and Myeloma cells. The observation of the cell morphology showed that there was an apoptotic process on the treated Myeloma cells, and not on the treated Raji cells. The result of the Immunocytochemistry assay showed that the C. mangga oil induced apoptosis on Myeloma cell lines by increasing expression of P53.
