Summary:ABSTRACT: An investigation has been performed to monitor cephalexin in blood, urine and feces of rats following administration of its derivative N-benzoyl-cephalexin. Female Wistar rats were randomly divided into two groups of six animals. To the first group N-benzoyl cephalexin (equivalent to cephalexin 25 mg/kg) was given orally in 10% PVP suspension to the fasted rats. Blood samples (200141 were taken periodically at 0, 2, 10, 30, 40, 50, 60, 90, 120, 150, and 180 minutes after drug administration from the lateral tail vein to analyze the unchanged cephalexin content fluorometrically (excitation 345 nm and emission 425 nm). The second group of the fasted animals were received the same treatment, after which urine and feces were collected during 0-30 hrs for the analysis of unchanged cephalexin. The results have shown that within 0-6 and 6-12 hours, the respective amounts of urinary excretion of cephalexin were 4.85 ± 2.79 pg and 5.58 ± 2.99 pg, with total amount excreted during 0-30 hours of 23.52 ± 11.86 pg. There was no N-benzoyl moiety was found in the urine after TLC, but only cephalexin, indicating that all the absorbed derivative was biotransformed into its parent compound. The amount of cephalexin found in feces for 30 hours collection was 201.4 1- 58.3 jig, or 5.90% of the cephalexin dose. Keywords : Cephalexin, N-Benzoyl cephalexin, Pharmacokinetics
