Summary:Dental plaque may cause tooth caries and other oral diseases. It contents bacteria bound to the matrix of salivary glycoproteins and extra-cellular polysaccharide. Dental plaque may be controlled chemically by mouth rinsing. Papain is a proteolysis enzyme which capability to catalyze peptide bound to simple chemical compounds. The purpose of this study was to determine the effect of mouth rinsing with papain enzyme solution 0.1% and 0.2% to the colony of Alpha Streptococcus and the growth of dental plaque in 13 � 15 years old girls. The subjects comprise of 23 girls divided into 3 groups. Each group rinsed their mouth with different concentration of papain enzyme solution, namely 0% (control group), 0,1% and 0,2%, once a day after brushed their teeth at night before sleep for two weeks. The data were taken just got up their sleep at early morning before eating and brushing. It is done in 3 times, namely before, after one week and after two week rinsing with papain enzyme solution. The data of Alpha Streptococcus colony was taken by swabbing with sterile cotton bud at the buccal surface upper molars then inoculated in blood growth media and data of the growth of dental plaque by scoring the plaque with PHP method modified by Amith. The data was analyzed by Kruskal Wallis, Mann Whitney and Spearman correlation at the 0,05% significant level. The result showed that there was significant difference of the colony numbers decreased of Alpha Streptococcus after rinsed papain enzyme solution for two weeks (�² = 9,63
