prostaglandin syntesis prior to ovulation, and the cyclooxygenase-2 (COX-2) enzyme is a key
enzyme that important in biosynthesis of prostaglandin. LH direct activates the cyclic adenosine
monophosphat dependent protein kinase (PKA) pathway, were equally able to up regulate COX-
2 expression in pre ovulatory granulosa cells. Curcumin is a natural polyphenol with various
beneficial biological activities and was shown to be a potent inhibitor of COX-2.
The objective of this study is to asses the inhibition of curcumin on COX-2 expression in
granulosa cells of the ovarian follicle and to know the action site of curcumin on signal
transduction in cAMP pathway.
The subject of this study were 27 female rattus norvegicus strain sprague dawley. At the age of
28 the subject were induced by 5 IU PMSG (Pregnant Mare Serum Gonadotropin). Rats were
stimulated by 5 IU LH (Luteinizing Hormone),then administered by teophylline 25 mg/kgBW
orally and curcumin 100mg/kgBW orally at 24 hours after PMSG administration. Aquades was
administered as a control group. Rats were sacrified at 30 days of age. Evaluation of COX-2
expression and immunohistology samples of granulosa cells of ovary used
immunohistochemistry technique with prime Antibody of Rabbit Monoclonal (RB-9072-P) to
determine the antigen location.
The result showed that COX-2 expression in curcumin group significantly decrease than in
control group and COX-2 expression in LH+theophylline+curcumin groups significantly
decrease than in LH+theophylline groups.
The conclusion of this study suggest curcumin inhibited COX-2 in granulosa cells ovary follicle
and the action site of curcumin inhibition is in the upstream of cAMP in protein kinase (PKA)
pathway. |
