The two CYP2C subfamilies, these are CYP2C9 and CYP2C19, exhibit wide inter-individual variation in expression which influence the drug metabolism capacity. Indonesia as an archipelago, consist of hundreds different ethnic groups. One of the Indonesian ethnic origin is Buginese population in Makassar, South Sulawesi. The interest variant allele of CYP2C9 in this research is CYP2C9*3. This allele have reduced catalytic activity compared to that of wild type CYP2C9*1. It influences the decrease of clearance of parent drug and increase the toxicity risk of the drug. Whereas, the interest variant allele of CYP2C19 in this research is CYP2C19*17. This allele leads to an increased rate of transcription and increased metabolism of the drug compared to that of wild type CYP2C19*1. Its consequences are the lack of response of the parent drug and low terapeutic effect of the drug. The aims of this research are to determine the frequencies of the CYP2C9 genotype, which contain *1 and *3 allele variant, also the CYP2C19 genotype, which contain *1 and *17 allele variant in Buginese population, which was divided into male and female subpopulation. Ninety six Buginese subjects, consist of fourty eight male subjects and fourty eight female subjects of Buginese origin were studied. CYP2C9 and CYP2C19 alleles were detected by PCR-RFLP assay. The result showed the frequencies of CYP2C19*3 and CYP2C19*17 allele on overall population of Buginese origin were 1.56 % and 4.68 %, respectively. The frequencies of CYP2C19*3 allele on female subpopulation (2.08 %) was insignificantly higher than male subpopulation (1.04 %), whereas the frequencies of CYP2C19*17 allele on female subpopulation (8.33 %) was significantly higher than male subpopulation (1.04 %). None of the subject had the CYP2C9*3/CYP2C9*3, CYP2C19*17/CYP2C19*17, or CYP2C9*3/CYP2C19*17 genotypes on ninety six subject of Buginese population.
