Aims.This study purposed to identify epitopes of P.gingivalisin peripheral blood serum of ACSpatients. Methods. Twenty two ACSpatients were enrolled in this study and Western blottingmethod was used to identifyP.l1#Jingivalis epitopes. Briefly,Pgingivalis antigen was fractionated by SDS-PAGEand then transferred into nitrocellulose membrane. Antigen fractions were then reacted with primary antibody (polyclonal antibody from peripheral blood serum of ACS patients) and secondary antibody (anti human IgG-biotin). Epitopes of P.gingivaliswere identified in membrane blot after being reacted with chromogenicenzyme SA-HRPand substrate TMB.Results. Allof ACSpatients beared P.flgingivalis epitopes in their blood. Amount of epitopes varied, range between 3 to 20 epitopes in each patient. Conclusion and suggestion. The existence of P.gingivalis epitopes in peripheral blood serum of ACSpatients suggested the role of Pgingivalisin ACS. Further studies were needed to elucidate pathomechanism of each epitope in ACS.P.r?gingivalisepitopes might be useful as target molecules for generating monoclonal antibody which could be a tool for predicting and preventing ACS</p> |
