Langkah Sembiring, M.Sc., Ph.D. ETD A research on exploration of rhizosphere streptomycetes producing antifungal on various plants (Cyperus rotundus L., Zea mays L., Rhizopora mucronanata dan Rhizopora apiculata) was conducted. Seventeen isolates of streptomycetes was tested to asses their potency in producing antifungal compound by using agar block method with Aspergillus niger FNCC 0060, A. terreus FNCC 0061, A. flavus NRRL 3357, Candida albicans ATCC 10231, C. albicans ATCC 24433, Tricoderma reesei NBRC 31329, Fusarium oxysporum KFCC 11363P, and Trichophyton consentricum A11 as test fungi. The result showed that 10 isolates among 17 isolates showed their potency to produce antifungal. Based on their antifungal activity, 2 isolates (J10 dan 41X) were chosen to produce antifungal compound using fermentation method in GYE broth. The medium then was extracted by using etil asetat. Characterization of antifungal compound was carried out by using TLC and bioautography test. The result of bioautography test showed that the spot from TLC of J10 and 41X isolates could inhibit C. albicans ATCC 10231 dan T. reesei NBRC 31329 growth. Identification of antifungal compoud of J10 and 41 isolated by using TLC analysis and spray reagent sugessted that then alkaloid, flavonoid, and phenol class of compound. Therefore, identification with GC-MS, the compounds were not be able to determined. Characteritation and identification of isolates were based on colour grouping, gram staining, morphology spora chain and spora surface ornamentation. The result showed that J10 and 41X isolates were gram positif bacterial, types of morphology spore chains were spiral to J10 and flexus to isolate 41X, and spora surface ornamentation were both spiny. Based on their spora chain morphology and spora surface ornamentation, isolate J10 was putatively identified to be member of Streptomyces novaecaesareae and isolate 41X to be member of S. litmocidini. Therefore it could be concluded that rhizosphere streptomycetes isolated from various plants were potencial to produce antifungal compound.
