Summary:On the effort to obtain a carbon source and fermentation conditions that most suitable for cellulose production by Acetobacter lovaniensis strains (MGA 6, SLK 1) and Gluconobacter oxidans (GDN 32) were performed by static method. Optimation of carbon source were done by a variety of fruit juice (pineapple, manggo, orange, and papaya) and various pure sugars (glucose, fructose, and sucrose). Based on the best carbon source and strain optimation of fermentation conditions which interms concentration of carbon source, volume of inoculum, pH, and incubation temperature. Production of cellulose was carried out at the optimum conditions and the productivity of cellulose was determined by the yield value. Cellulose produced were identified by the method of FTIR spectroscopy and the determination of the cellulose quality by the method of X-ray difractometry. The results showed that the best carbon sources were pineapple juice for MGA 6 strain and sucrose for SLK 1 strain. Pineapple juice with the optimum fermentation conditions was that the concentration of carbon source with a ratio of fruit : water was 1:1 , 10 ml of inoculum volume, 6 of pH the medium, and 30oC at incubation temperature, while 3% (w/v) of sucrose concentrations, 10 ml of inoculums volume, 5 of pH the medium, and 30oC at incubation temperature. Productivity of bacterial cellulose with pineapple and sucrose 4.41% and 26.12%. Quality of cellulose is related to the tensile strength fiber of cellulose which determined by crystallinity index. Crystallinity index of pineapple by static and agitated method was 83.04% and 93.65%, while sucrose 91.92% and 65.35%. Therefore, pineapple with agitated has higher tensile strength fiber than sucrose.
