Summary:Congenital toxoplasmosis is a disease caused by Toxoplasma gondii. It can infect fetus intra uterine. Toxoplasma gondii causes heavy infection in congenitally infected children. Polymerase Chain Reaction as a diagnostic tool has not been known well and familiarly used in Indonesia. Therefore, implicating this technique requires a lot of research. Quality and sensitivity of PCR method very depend on the gene target used. Repetitive sequence R522 in T. gondii genome has 300 copies therefore it is a great candidate to diagnose congenital toxoplasmosis. This research aims to observe the application of PCR targeting repetitive sequence R522 using maternal blood and amniotic fluid from pregnant mice. This research used mated female mice. Mice were infected with 1x10 3 T. gondii tachyzoites. Maternal blood, amniotic fluid and fetus tissue samples were obtained to isolate DNA using DNAeasy Blood and Tissue (Qiagen) and tested by PCR using repetitive sequence R522 (C1-S1 and C1-S2) primer and primer targeting B1 gene. PCR results were analyzed descriptively. The results showed that primer arranged from repetitive sequence R522 by PCR method could detect T. gondii in infected mice. C1-S1 primer gave a better result by PCR method than with C1-S2 and B1 primers. Amniotic fluid sample is more suitable than maternal blood for diagnosing congenital toxoplasmosis.
