Summary:Background. Indonesia is a country in Southeast Asia consisting of many islands, both large and small islands. Residents in Indonesia consists of various tribes and have cultural and linguistic diversity. Generally in Indonesia consists of Malay ethnic in western and central area, and Melanesian ethnic in eastern Indonesia. PCOS is an endocrine disorder characterized byhyperandrogenaemia, anovulatoar, infertility andhirsutism with the prevalence in the population between 5-10 %. Gena CYP11a/ cytokrome P450scc located on the long arm of chromosome 15 at 15q23, is a prime importance gene in demonstrating the incidence of PCOS, whereas gene CYP19 / cytokrome P450 aromatase located on the long arm of chromosome 15 position 15q21.2 is a candidate gene supporting the occurance of PCOS. Aim. To determ ine the association of Polymorphism pentanukleotida (tttta)n Gena CYP11a with Testosteron serum level and gene Polymorphism CYP19 with Homeostasis model assesment (HOMA) inpatients with Polycystic Ovari Syndrome (PCOS) Melanesian and Malay ethnic. Location. RSUP dr. Sardjito Yogyakarta and RSU Jayapura Period : May 2008 until May 2011. Research type: Cross sectional study. Object: Melanesian and Malay ethnic with PCOS (n1=32, n2=30) and Melanesian and Malay ethnic normal women as a control (n3=30, n4=30). Intervention: 10 cc of peripheral blood taken for hormonal, GDP, Insulin, PCR and Gene Scan examination. Result: The result of statistical analysis found significant differences between short allel and long allel on CYP 11a (P=0,010) in PCOS and normal Melanesian and Malay women. On examination of CYP 19 polymorfisms found in PCOS Melanesian ethnic women, analysis results are not statistically significant differences (P=0,484). The results indicated: (1) Prevalence of Polymorphism > 5% both on Melanesian and Malay ethnic. (2) Characteristics of the hormonal analysis results in both ethnic Melanesia and Malay, are significant differences between hormonal LH, FSH , E2, P, but in T there is non significant difference. 3a) There is the influence of polymorphism gene CYP 11a and polymorphism gene CYP 19 on the hormonal profile in patients PCOS Melanesian and Malay ethnic. (3b) There is no correlation polymorphism gene CYP 19 wiyh the insulin resistance based on HOMA in patients with PCOS both Melanesia and Malay ethnic. (3c) There is significant difference of polymorphism gene CYP 11a at short allel (4R) and long allel (6R) both between groups PCOS and control groups on ethnic Melanesia and Malay. Conclusion: Polymorphism pentanukleotida gene CYP11a and CYP 19 has the role of hormone profiles in patients with PCOS in Melanesia and Malay ethnic.
