Summary:Cymbidium mosaic virus (CymMV) and Odontoglossum ringspot virus (ORSV) are common viruses infecting orchids. CymMV causes withered orchid, flower size become smaller, necrotic, and change the phenotype of flower. While ORSV causes color breaking and flower distortion. The infected orchid will lose vigor, reduce flowering and decrease quality of commercial value. Various methods to detect CymMV and ORSV have been widely applied. Neverthless, the molecular detection of both viruses is still rare. This study aims to determine orchids infected CymMV and ORSV and also to detect virus using the Multiplex RT-PCR method. In first stage, RNA was isolated from virus-infected leaf tissue. The positive samples then analyzed by Multiplex RT-PCR to detect both of viruses. The result showed positive CymMV at 669 bp and ORSV at 474 bp infected Phalaenopsis sp, Cattleya sp, dan Phaius tankertile using simplex and multiplex method with a mixture of both RNA viruses. Positive multiplex also shown with a mixture of their cDNA product.
