Summary: | This research is set in the eastern part of Kelantan State, which is Lata Janggut. As there is limited of the information on the diversity of microorganisms on Lata Janggut, the study was conducted to identify the microbes present in this freshwater ecosystem. The aim of this study was to conduct the Polymerase Chain Reaction (PCR) method by using 16S rRNA gene region and then to identify the microbes isolated from Lata Janggut using 16S rRNA gene region. There are total 15 pure colonies that were isolated and the isolated DNA from these pure cultures was then used for l6S rDNA sequence analysis. The isolates were obtained from the upstream and along the river bank of Lata Janggut. Four of the purified l6S rDNA products were sent for sequencing and the sequences obtained were used for bacteria identification through BLAST. The results obtained indicated that the isolated bacteria belonged to genus Aeromonas and the phylogeny trees were constructed using the gene sequences to confirm these results. The river was considered as moderately clean based on the Aeromonas spp. isolates which were found to be dominant in the water samples collected from Lata Janggut, Kelantan. In the nutshell, more studies such as biochemical test have to be done on Lata Janggut, Kelantan to determine the health of this popular waterfall. |
