Cellulase can be produced using fungi such as Aspergillus niger on solid state fermentation (SSF). But, the problem is the cost of producing the enzyme is expensive. The growing approaches to reduce the cost of cellulases production can be achieved via using of cheap, renewable agriculture wastes which consider an excellent carbon source for microbial enzymes production and also looking for microorganism having the ability to hyper-production of cellulases. Therefore, considering the advantages of SSF, the optimal chemical condition for the production of cellulase enzyme by Apergillus niger from solid-state fermentation of sugarcane bagasse is investigated. Three parameters was chosen to be screened and optimized using Design Expert Software (DOE). The parameters are amount of substrate (C-source), concentration of yeast extract (N-source) and concentration of Potassium di-Hydrogen Phosphate (KH2PO4) (Phosphate source). After screening, the parameter for N-source was not significant to the model. Thus, parameters C-source and Phosphate source was optimize.
