Phylogeography of bornean frog, limnonectes kuhli as inferred by mithochondrial DNA (mtDNA) of cytochrome c oxidase I (COI)

Limnonectes kuhli is a species of anuran taxonomically categorized under the family of Ranidae or also known by its common name of Kuhl's Creek frog or fanged frog. However, this taxonomic categorization proposed by Dubois (1987, 1992) is somewhat contradicted by research and claims made by Eme...

However, this taxonomic categorization proposed by Dubois (1987, 1992) is somewhat contradicted by research and claims made by Emerson and Berringan (1993) that the fanged frog were a monophyletic group, recognized at least two main species groups within the clade. Therefore, with the diverse representation of this species at different altitudinal gradience and nich populations displaying varying characteristics, genetic variation was examined between samples taken from two different sample locations; Limnonectes kuhli from Matang (Kuching, Sarawak) and Limnonectes kuhli from Gading (Kuching, Sarawak). Overall 6 samples were isolated: 4 from Matang and 2 from Gading. Genetic variations among samples were examined using PCR and DNA sequence analysis of Cytochrome C Oxidase I (COl) mtDNA gene. Unfortunately, DNA sequence analysis of samples isolated from PCR was unsuccessful rendering further study of phylogenetic analysis between the two separate populations inconclusive. Therefore, more samples from different populations and further optimization of PCR techniques are required to achieve the desired results of this study.
