Isolation, purification, and characterization of DNA from capsicum species (Capsicum annuum var longum) found in the Philippines

There are various medical purposes that had been found to be possessed by Capsicum species. In this study, the DNA from Siling Haba or Louisiana Long Pepper (Capsicum annuum var longum) found in the Philippines was isolated. The protocol utilizes fresh leaves sample from Pedefco Social Farm located at Malaining Bago Naic, Cavite, Philippines. These leaves were grounded with mortar and pestle, several plant cell contaminants such as polysaccharide and polyphenolic compounds were present which prevents the proper isolation and purification of the plant DNA. These contaminants were reduced with the presence of extraction buffer, PVP, and mercaptoethanol. DNA samples were then divided into two groups one containing RNase and the other without. Characterization of the DNA were made by introducing restriction enzymes such as EcoRI, HindIII and the mixture of both (EcoRI/HindII). These fragments were separated using agarose gel electrophoresis and viewed using ethidium bromide stain with the aid of a UV-transilluminator and were properly documented using a digital camera.
