Cytotoxic isothiocyanates from Moringa oleifera Lam seeds
The seeds of Moringa oleifera Lam were freeze-dried and extracts were obtained using dichloromethane. The extract afforded 4-(4-O-acetyl-a-L-rhamnosyloxy)benzyl isothiocyanate (MOS1), 4-(a-L-rhamnosyloxy)benzyl isothiocyanate (MOS8), squalene (MOS3), sitosterol (MOS2), triacylglycerol and long chain...

Summary: | The seeds of Moringa oleifera Lam were freeze-dried and extracts were obtained using dichloromethane. The extract afforded 4-(4-O-acetyl-a-L-rhamnosyloxy)benzyl isothiocyanate (MOS1), 4-(a-L-rhamnosyloxy)benzyl isothiocyanate (MOS8), squalene (MOS3), sitosterol (MOS2), triacylglycerol and long chain unsaturated hydrocarbons. The structures of MOS1 and MOS8 were elucidated by extensive 1D and 2D NMR spectroscopy and structures of MOS3 and MOS2 were identified by comparison of their 13C NMR data and 1H NMR data with those reported in literature. The cytotoxicity of MOS1 and MOS8 were evaluated against the human cancer cell lines, non-small cell lung adenocarcinoma (A549), colon carcinoma (HCT 116), and the non-cancer cell line Chinese hamster ovary cells (AA8) using the MTT cytotoxicity assay and compared with Doxorubicin as positive control. Compound MOS1 gave IC50 values of 10, 12 and 22 µM and compound MOS8 gave IC50 values of 12, 11 and 15 µM for A549, HCT 116 and AA8, respectively. Doxorubicin gave IC50 values of 4, 3 and 4 µM, respectively. Results revealed that MOS1 and MOS8 were cytotoxic against the different cancer cell lines tested. The leaves of M. oleifera were air-dried and extracts were obtained using dichloromethane. The extract afforded polyprenol (MO7), phytyl fatty acid ester (MO10), lutein (MO6) and long chain unsaturated hydrocarbons. The structures of MO7, MO10 and MO6 were also obtained based from comparison with those reported in literature. |
