Summary: | This thesis explores the development of a novel disposable immunosensor based on Fe2O3 core with carbon paste and gold coating. The project explores the preparation, characterisation and measurement of the various properties of the immunosensor which will be helpful in the detection of Prostate-Specific Antigen (PSA). The detection of PSA is done by an amperometric route with a constant voltage of -300mV vs. Saturated Calomel Electrode (SCE).
The combination of Fe2O3 core, carbon paste and gold coating showed good adsorption properties for the anti-PSA (antibody) and yielded very good results. 5μl of 5 mg/ml anti-PSA is immobilised onto the electrode by physical adsorption for a time period of 24 hours overnight at 4oC. The electrode was tested against antigen at concentrations of 0.25 mg/L, 0.5 mg/L and 0.75 mg/L in pH 7.4 Phosphate Buffer Saline (0.1M). It was observed that the current increased by a substantial value of 3 orders, and this presents us with a good electrochemical response for the detection of PSA. This methodology can thus be proposed for use in clinical applications and even be used for detection of other antigens. |
