The secretome with lignocelluloses hydrolyzing enzymes by two strains of Trichoderma reesei grown on cellulose, starch and agricultural waste such as saw dust and corn stover were analyzed using a proteomics approach with high throughput isobaric tag for relative and absolute quantification (iTRAQ) technique using liquid chromatography-tandem mass spectrometry (LC-MS/MS). The iTRAQ quantification of the secretory proteins of T. reesei showed significant up-regulation of different cellulose hydrolyzing endocellulases, exocellulases, β-glucosidases and glycoside hydrolases and hemicellulases including endo-1,4-beta-xylanase, beta-xylosidase, alpha-glucuronidase, alpha-L-arabinofuranosidase, beta-mannanase, beta-mannosidases polygalacturonase, glucuronidases and arabinosidases. The lignin degrading enzymes (peroxidases, laccases, glyoxal oxidases, catalase, cytochrome-c oxidase, and superoxide dismutase), proteases, protein translocating transporter proteins and several novel proteins were also identified in the secretome.
