This project investigates the interaction between adipocytes and other endocrine cells with the main focus on NPY. We aim to better understand the mechanisms of NPY secretion so that the NPY network can be manipulated to combat the growing problem of obesity in the world. Results obtained showed that NPY is compartmentalized into numerous vesicles, which means NPY is secreted through the endosomal system. Substances which induced the secretion of adipokines also aided in the trafficking of the adipokines’ vesicles. This meant that NPY can also be released via regulated exocytosis of its vesicles. Experimental results also showed that TNF-a and leptin enhanced intracellular expression and NPY secretion, while epinephrine lowered the NPY expression and secretion. It was also inferred that NPY has constitutive and regulated secretions. In addition, NPY is transported through the Golgi apparatus. Lastly, regulated secretion of NPY is calcium dependent whereas constitutive secretion of NPY is calcium independent.
