Summary:In this thesis, by experimental exploitation and mechanism investigation, we endeavour to finally develop and apply an innovative living tissue based scaffolding system (LS) that is completely free of scaffold material components to enable phenotypically bona fide pure neo-tissue formation for artificial graft per se ultimately. Technically, the implementation process involves: 1) Novel “edge flourish” (EF) inspired phase transfer cell culture (PTCC) in micro-cavitary hydrogel (MCG) scaffold propelling neo-tissue islets formation to fill up the created vacancy and further expand throughout the whole hydrogel; 2) LS formation and hydrogel removal inducing macroscopic framework of a LS that maintains the overall shape and size during removal of hydrogel components by a gentle dissolving treatment. Successful fabrication of LS via newly PTCC strategy, as a porous scaffold made of pure and living cartilaginous tissue, can be further explored and applied to develop macroscopic true cartilage grafts. It can also be employed as a fine model for manipulation of endochondral osteogenesis and osteogenic angiogenesis.
