In this project, the fabrication of PDMS (Polydimethylsiloxane) gradient generator and experiments of concentration gradient generation was carried out. The concentration gradient generator is a microfluidic device that was designed to generate chemical gradients of singular or dual chemical species; chemical gradient generation will be potentially useful in analyzing the spatial and temporal behavior of organisms under the influence of various concentrations of single of dual chemical species. This project mainly focused on creating a bubble-free environment to observe the diffusion of florescence (FITC). Several chips designs were tested for the effective generation of gradients. The general design of the devices included fluidic pressure balance between 4 fluidic sources with 2 sinks, and 4 fluidic sources with 4 sinks. Furthermore, a circular gradient generator with 4 fluidic sources and 2 sinks was also tested.
