We have developed a novel hydroxyl radical footprinting (HRF) technique for studying in vivo structural dynamic in membrane proteins in their native biological states on the cell surface. We have successfully applied the technique to elucidate the voltage gating mechanism of porin, an integral membrane protein, of E coli directly on the living cell. We further extended the method to study in-vivo structure dynamic of integrin aLp2 heterodimer, a pair of important membrane proteins of mammalian cell related to many human diseases.
