Biomaterial surface engineering plays a pivotal role in the design and development of various biomedical devices and engineered tissue equivalents. The modification of tissue culture substrate with thermo-responsive polymer (TRP) has led to the realization of the enzyme-free harvesting of anchorage-dependent cells through an acute increase in surface hydrophilicity of TRP across its lower critical solution temperature (LCST). To date, the intricate mechanisms of cell deadhesion/adhesion on TRP and other bio-functionalized polymer have not been fully investigated. The understanding of biophysical behavior of cells towards the thermo-physical stimulations and biological recognition on biomaterials will be critical to the design of highly tailored cell regeneration processes. In my thesis, highly integrative biophysical approaches are applied to develop quantitative correlation between the biophysics of cell adhesion/deadhesion, receptor-ligand interactions, cytoskeleton reorganization and biomaterial properties.
