Summary: | This paper reviews results of experiments conducted to understand how the liposome permeability across the lipid bilayer is affected, after which liposome formulation with the best retention capabilities was achieved. Fluorescence spectrometry test was performed on liposome samples composed of 15:0 PC lipids which encapsulated 5(6)-Carboxy-fluorescein dye over a period of one month. The encapsulated dye serves as a marker for amount of content leakage over time.
To further substantiate the results, mean size distribution of the samples were performed with dynamic light scattering zeta-sizer to characterize the initial mean size particle distribution and changes of the sample over the time-study period.
In addition, the emission intensity change of the dye encapsulated liposome was being fitted into the Fick’s Model based upon the Fick’s First Law of Diffusion, subsequently obtaining the first-order rate constants and permeability coefficient of the samples. With that a comparison of how well the liposome formulation is able to retain the encapsulant was made with that of literature. |
