Summary:In this project, we develop an image analysis algorithm to work with a surface plasmon resonance (SPR) biosensor for multiple bio-molecular interactions detection. The SPR biosensor measures the color change produced by surface plasmon excitation in the spectral domain. When bio-molecular interaction occurs at the sensor surface (such as antibody-antigen interaction), the refractive index value is varied and corresponding spectral shift will be produced in the spectrum. In the experimental works, we demonstrated the performance of the system with the detection of different refractive index samples ranged from 1.3333 RIU to 1.3470 RIU. The system sensitivity is found to be 1.84 x 10-5 RIU. This value is nearly 3 times better than the resolution provided by existing spectral based SPR imaging sensor. The system was further applied for antigen-antibody bio-molecular interactions detection. The bio-molecular interaction between different concentrations of BSA antigen (10 mg/ml, 5mg/ml, 2.5 mg/ml and 1.25 mg/ml) and BSA antibody molecules have been measured. The results of this project prove the capability of the SPR biosensor applied for bio-sensing application (antibody-antigen interaction). In future work, the SPR biosensor can find potential applications in biomedical research for the detection of different bio-molecular interactions in the laboratory.
