Ever since the discovery of microscopes through stacking a pair of lenses by Anthony Leeuwenhock, bio-imaging and bio-sensing have been the two important areas which rapidly developed in medical sciences research. The microscopes enabled viewing of many micro-organisms in water and this has increased the quest among researchers to explore different techniques in microscopy that can aid individual cell monitoring. Analyzing and observing individual cells is necessary for many clinical applications (e.g., detection of cancer). However, as many biological samples (e.g., cells) exhibit low contrast, the traditional bright field microscopes could not provide sufficient contrast to view these transparent samples and this led to the discovery of many microscopic techniques such as phase contrast microscope and fluorescent microscopes etc. Now, fluorescent microscopy has been an established technique for individual cell monitoring. Even though fluorescent techniques are widely used, they suffer from certain drawbacks such as cyto-toxicity due to exogenous labeling, sample preparation time and photo-bleaching effect. These disadvantages inhibit longer term continuous individual cell monitoring during assays. This thesis attempts to overcome these limitations by applying thermal lens response and imaging system, an alternative microscopic sensing technique, for real-time, non-invasive individual cell monitoring.
