Oxidative stress due to overproduction of ROS is a key contributor to pathogenicity of obesity and its related metabolic disorders. Adipose tissue is recognized as a complex organ consists of various cell types including adipocytes and macrophages. Macrophages in adipose tissue are major source of ROS found in obesity. Apelin is one of adipokines secreted by adipose tissue and known for anti-diabetic and anti-obesity properties. Plasma apelin has been identified as circulation hormones and both apelin level and oxidative stress are increased in obesity. However, role of apelin regulation on ROS production in macrophages remain unknown. In present study, we provide the evidence that apelin supresses ROS production in macrophages. We also observed that apelin is able to abolish the TNFα -induced dysregulation of anti- and pro-oxidant enzymes expression in macrophages. In addition, we further observed that apelin promotes anti-oxidant enzymes through PI3K /Akt pathway, and suppresses the expression of pro-oxidant enzymes via MAPK/ERK pathway. This study reveals that anti-oxidant properties of apelin in macrophages, and suggests its potential application as a novel therapeutic target for metabolic diseases.
