Summary: | A novel algae DNA extraction protocol was developed after testing with three QIAGEN™ extraction kits (PS, PB, PP) on two types of algae species L. variegate and H. caespitosa. The mean and standard deviation (S.D.) values were tabulated based on the Nanodrop and Qubit results and grouped by extraction kits or preparation methods (FF/FD). Gel images were correlated to Qubit readings post-PCR to confirm the DNA concentration measured. Overall, PS was the better option due to higher DNA quality and consistent PCR results using FF L. variegate and FD H. caespitosa samples. In conclusion, the novel extraction protocol could be used for sequencing and identification of bacterial community within the algae species. |
