Summary:This research uses cell-line L929, a fibroblast which is a type of cell present in animal tissues that has structural framework and acts as a connective tissue. The aim of this study is to ascertain a fundamental understanding on the decellularisation process involving surfactants and to quantify the effect of the surfactant on adherent cells using Quantitative Phase Interferometry (QPI). The surfactant used for this study is Triton X-100. Different concentrations of Triton X-100 were added to samples of L929 cells and observations for cell detachment were done using Light Microscope and a QPI set-up. Images of cells during each process through the QPI set-up were then quantified and analysed using QPI and a MATLAB programme. It was apparent that there was minimal but distinct effect on the adherent cells by Triton X-100. It was observed and quantified that Triton X-100 consistently caused the cells to shrink in size and flatten but did not cause the cells to detach.
