Summary:Genetic relationships among 14 longan cultivars were investigated using the high annealing temperature RAPD (HAT-RAPD) technique. Eighteen decamer-arbitrary primers produced reliable DNA polymorphism ranging in molecular weight from 100 to 2,500 bp. A total of 214 polymorphic loci were analyzed by cluster analysis and UPGMA to present a dendrogram depicting the degree of genetic relationships among the 14 longan cultivars. Our result indicated that all cultivars were classified into two major groups. The first group was divided into two sub-groups and the second contained five sub-groups, while the third, forth and fifth sub-groups consisted of different single members: 'Bai Dam', 'Hokkian' and 'Sai Namphueng'. In addition, somatic hybrids between different longan cultivars, 'Do' x 'Bai Dam', 'Do' x 'Biao Khiao','Biao Khiao' x 'Bai Dam' and their reciprocals could also be detected by the HAT-RAPD technique. © ISHS.
