The Journal is published 4 issues/year (i.e., Mar, Jun, Sep, and Dec). วัตถุประสงค เพื่อศึกษาผลของสารสกัดสมุนไพรพรมมิที่มีตอโปรตีน Tau ในเซลล PC12 ที่ไมไดรับ nerve growth factor (NGF) วิธีการ นำเซลล PC12 ที่ไมไดรับ NGF มาเลี้ยงในอาหารที่มีซีรัมปกตินาน 1 สัปดาหกอนเปลี่ยนมาเลี้ยงในอาหารที่มี ซีรัมต่ำนาน 1 สัปดาห จากนั้นแบงเซลลเปน 2 กลุม กลุมแรกเลี้ยง 7 วันเพื่อใชสำหรับการวิเคราะห 3-(4,5Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) กลุมที่สองเลี้ยงในอาหารที่มีซีรัมปกติหรือที่ปราศจากซีรัม ที่ไมมีสารสกัดพรมมิหรือมีสารสกัดพรมมิความเขมขน 50 ถึง 300 μg/mL นาน 2 วันเพื่อใชสำหรับการวิเคาระห immunoblot สำหรับการวิเคาระห MTTในแตละวันคาเฉลี่ยของความรอดชีวิตของเซลลที่เลี้ยงในอาหารทมี่ีสารสกัดพรมมิแตละความเขม ขนถูกนำมาเทียบกับความรอดชวีิตของเซลลที่เลี้ยงในอาหารที่ไมมีสารสกัดพรมมิ การทำ immunoblot โดยใชแอนติบอดีตอ โปรตีน Tau โปรตีน dephosphorylated Tau และ glyceraldehyde dehydrogenase และวิเคราะหปริมาณโปรตีนดวยโปรแกรม Scion Image แลวนำมาคำนวณคา Tau-1 immunoreactivity ผลการทดลองทั้งหมดวิเคราะหโดยใช one-way ANOVA ผลการทดลอง สารสกัดพรมมิความเขมขน150 และ 200 μg/mL เพิ่มความรอดชีวิตของเซลล PC12 ที่ไมไดรับ NGF ที่เลี้ยง ในอาหารที่มีซีรัมปกตินาน 4 วันอยางมีนัยสำคัญขณะที่สารสกัดพรมมิความเขมขน 50 μg/mL มีแนวโนมที่สามารถลดความ รอดชีวิตของเซลล PC12 ที่ไมไดรับ NGF ที่เลี้ยงในอาหารที่ไมมีซีรัม นาน 2-4 วัน สารสกัดพรมมิความเขมขน 100 μg/mL สามารถลดทั้งปริมาณรวมของโปรตีน Tau และปริมาณ phosphorylated Tau ในเซลล PC12 ที่ไมไดรับ NGF ทั้งที่เลี้ยงใน อาหารที่มีซีรัมและในอาหารที่ปราศจากซีรัม นอกจากนี้สารสกัดพรมมิสามารถเพิ่ม Tau-1 immunoreactivity ในเซลล PC12 ที่ไมไดรับ NGF ทั้งที่เลี้ยงในอาหารที่มีซีรัมในลักษณะ dose-dependence สรุปผลการทดลอง สารสกัดพรมมิสามารถเพิ่มความรอดชีวิตของเซลล PC12 ที่ไมไดรับ NGF และสามารถลดทั้งปริมาณ รวมของโปรตีน Tau (Tau 5) และปริมาณ phosphorylated Tau (ที่ตำแหนง Tau-1) ในเซลล PC12 ที่ไมไดรับ NGFที่เลี้ยงใน อาหารที่มีซีรัมและในอาหารที่ปราศจากซีรัม 2019-05-07T10:02:42Z 2019-05-07T10:02:42Z 2012 บทความวารสาร 0125-5983 https://www.tci-thaijo.org/index.php/CMMJ-MedCMJ/article/view/87845/69262 http://cmuir.cmu.ac.th/jspui/handle/6653943832/65137 Eng Faculty of Medicine, Chiang Mai University
