Summary: | Carpenter bee is one of pollinator which shows high potential and energetic for pollination. The gut microbiota especially intestinal bacterial may play important roles on host heath. The aim of this study was to investigate intestinal bacteria diversity via both approaches including culture-dependent and culture-independent (illumine solexa). Six carpenter bees ware collected and identified as Xylocopa latipes by using insect morphology and COI gene. For results of culture-dependent, total bacteria of fore gut, mid gut and hind gut were 7.14±1.79, 6.06±1.28 and 7.19±1.36 LogCFU/ml respectively. Total and Lactic acid bacteria in each section were not statistically different. All Lactic acid bacteria isolates cannot inhibit American foulbrood pathogen. In part of culture-independent, order Lactobacillaceae, Enterobacteriaceae, Carnobacteriaceae and Porphyromonadaceae were dominate bacteria in fore gut, family Lactobacillales, order Enterobacteriaceae, Carnobacteriaceae, Pseudomonadaceae, Porphyromonadaceae and Flavobacteriaceae were dominate in midgut. Lastly, in hind gut, dominate were similar to midgut but adjunct to Bifidobacterium sp. Alpha diversity Shannon and Simpson analysis showed that, foregut-midgut were not statistically different (P= 0.9362, 0.8102 respectively) while foregut-hindgut and midgut-hindgut were statistically different (Shannon P= 0.1307, 0.008239 respectively; Simpson P= 0.008239, 0.01307 respectively). Beta diversities use NMDS and PCoA, foregut-midgut and midgut-hindgut were not statistically different (P=0.3159, 0.0965 respectively) but foregut and hindgut were statistically different (P=0.002). this data can improve or predict their health carpenter bee |
