Summary:a-Amylase belongs to family 13 glycosyl hyrolase which cleaves a-1,4-glycosidic linkages of the starch into oligosaccharides, a-limit dextrin, and glucose. Amylases are used in a number of industrial processes, such as starch processing industry and textiles industry. Bacillus licheniformis a-amylase (bla) is one of thermostable enzyme which is widely used in starch liquefaction process. Bla has a thermostable property because it is activated by calcium–sodium–calcium metal triad and also has the third calcium resulting in more rigid structure. To meet the industrial need, the bla should be produced in large quantities. Pichia pastoris is one of expression system which is able to produce protein at high level. The aims of this research were to express gene encoding B. licheniformis SITH a-amylase (bla) in P. pastoris and to characterize the bla gene product.
