These nanobiosilica may be used as a matrix <br /> <br /> <br /> <br /> <br /> for gel filtration coloumn. In this present research, nanobiosilica of Thalassiosira sp. was <br /> <br /> <br /> <br /> <br /> studied as a matrix for separation coloumn. To obtain nanobiosilica of Thalassiosira sp., first <br /> <br /> <br /> <br /> <br /> lipid had to be extracted from the cell followed by acid treatment and calcination at 600oC <br /> <br /> <br /> <br /> <br /> with heat rate of 3oC/min for 6 hour. Surface topology and chemical composition of the <br /> <br /> <br /> <br /> <br /> nanobiosilica was characterized by scanning electron microscopy-energy dispersive <br /> <br /> <br /> <br /> <br /> spectroscopy (SEM-EDS). Nanobiosilica of Thalassiosira sp. was applied for matrices of gel <br /> <br /> <br /> <br /> <br /> filtration coloumn to separate mixtures of BSA and lisozyme. The results showed that one <br /> <br /> <br /> <br /> <br /> nanobiosilica was obtained from 100 g wet biomass. The topology of white nanobiosilica of <br /> <br /> <br /> <br /> <br /> Thalassiosira sp. has cylinder in shape with porous size of 80400 nm distributed randomly <br /> <br /> <br /> <br /> <br /> in the surface. The composition of SiO2 of Thalassiosira sp. cells before and after acid <br /> <br /> <br /> <br /> <br /> treatment was 10.8 and 93.79%, respectively. Separation of BSA and lisozim mixtures by <br /> <br /> <br /> <br /> <br /> chromatography coloumn based on a nanobiosilica matrix produced two protein fraction at <br /> <br /> <br /> <br /> <br /> nos. 1517 and 2021. By SDS-PAGE analysis, a protein band of 66 kDa of fraction no. <br /> <br /> <br /> <br /> <br /> 1517 was identified as BSA, but no protein band of fraction no. 2021 was observed due to <br /> <br /> <br /> <br /> <br /> a little amount of protein.
