The derivation of palm oil starts from the conversions of triglycerides to fatty acids which is hugely consumed by biodiesel-producing company or cosmetics and personal care company as feedstock. Production of fatty acids nowadays still uses Colgate-Emery process which react oil and water in 5000 kPa and 523 K, resulting in a high energy demand which is not environmentally friendly. In that case, an alternative process is very much needed. Enzymatic reaction which is commonly termed lypolysis is one of the alternative that can be used since the reaction is running under atmospheric temperature and pressure. Enzyme lipase used in lypolysis can be derived from microorganisms, and plants with plant’s derived lipase gives advantages in terms of the ease and cost to isolate the enzyme and also unrequirement to use enzyme in its concentrate forms so the cost can be further reduced. Studied showed that lipase from Plumeria sp. gives a high enzyme activity, hence it will be used in this research. In this research, the effects of parameter of lypolysis to the kinetic’s parameter is studied. Variable studied are substrate concentration (water volume fraction 2%, 4%, 6%, dan 8%), the amount of enzyme used (1%, 2%, and 3% of medium total mass), and mixing rate (variated in 680, 1230, 1520, and 1813 rpm). The increase in volume fraction caused the rate of reaction increases with an inhibition of substrates is showed in variation of 8% water. The research also showed product inhibition. The same trend is also showed when the enzyme and mixing rate used is increased. The result than is used to determine the parameters needed in reaction kinetic equation. <p align="justify"> <br /> <br /> text
