This study aims to increase in the level and productivity of lutein and zeaxanthin by increasing nitrate as a source of nitrogen in C. pyrenoidosa culture. In addition, an analysis of the effect of nitrate concentration on the growth kinetics and productivity of C. pyrenoidosa biomass was also carried out. The initial variation of NaNO3 given was 1 g/L (control) 1.5 g/L, 2 g/L, 2.5 g/L, and 3 g/L. Cultivation of C. pyrenoidosa culture lasted for 12 days. The culture was extracted using ethanol, KOH, and diethyl ether. Total lutein and zeaxanthin were then quantified using spectrophotometer. The results indicated that by increasing the concentration of NaNO3 up to 2 g/L in C. pyrenoidosa culture, lutein and zeaxanthin levels were increased by 101.1% and 106.1%, respectively, this followed by an increase in the productivity of lutein and zeaxanthin by 234.2% and 263.2%, respectively. The addition of NaNO3 concentration above 2 g/L can inhibit growth so that the production of lutein and zeaxanthin is also inhibited. text |
