<br />
as phenolic and flavonoid compounds can reduce and protect cells from damaging. Many plants <br />
<br />
contain phenolic and flavonoid compounds included pumpkin. The objectives of this research were <br />
<br />
to study antioxidant activity of flesh, peel, and seed of pumpkin by determining IC50 of DPPH (2,2- <br />
<br />
diphenyl-1-picrylhydrazyl) and EC50 of CUPRAC (Cupric Reducing Antioxidant Capacity); total <br />
<br />
phenolic and flavonoid content; analyze the correlation between total phenolic and flavonoid with <br />
<br />
their IC50 DPPH and EC50 FRAP, and analyze the correlation beetwen DPPH and FRAP in sample <br />
<br />
extracts. Extraction was performed by reflux using gradient polarity solvents. Determination of IC50 <br />
<br />
DPPH, CUPRAC EC50 capacity, total phenolic and flavonoid content of each extract were performed <br />
<br />
by spectrophotometry uv-visible and correlation of total phenolic and flavonoid content with their <br />
<br />
IC50 of DPPH and EC50 Đ» 􀀦􀁚!􀁗 ®ĤðŊðĤð±Ěŋ±Ė±  ĉ Ăő􀇌±¸ ÂÂő􀁗± ĖĚĐĉ􀍛ĚĈ±ĤÃÂи. N-hexane peel extract <br />
<br />
of pumpkin had the lowest IC50 DPPH (5.55 ± 0.10 µg/mL) and ethyl acetate seeds extract of <br />
<br />
pumpkin EC50 CUPRAC (104.17 ± 4.37 µg/mL ). The highest total phenolic content was given by the <br />
<br />
ethyl acetate peel extract of pumpkin (4.09 ± 0.10 g GAE/100 g), while the n-hexane flesh extract <br />
<br />
of pumpkin showed the highest total flavonoid content (11.22 ± 0.28 g QE/100 g). Total phenolic <br />
<br />
content in seed extracts of pumpkin had negative and significant correlation with their EC50 <br />
<br />
CUPRAC, while total flavonoid content in flesh, peel and seeds extracts of pumpkin had negative <br />
<br />
and significant correlation with their EC50 CUPRAC. IC50 DPPH of flesh and peel extracts of pumpkin <br />
<br />
had significantly positive correlation with their EC50 CUPRAC. All organs extract of pumpkin were <br />
<br />
categorized as very strong antioxidant activity by DPPH method, hence it was potential to be <br />
<br />
developed as source of natural antioxidant. Phenolic compounds were the major contributors in <br />
<br />
antioxidant activity of seeds extracts of pumpkin by CUPRAC method. Flavonoid compounds were <br />
<br />
the major contributors in antioxidant activity of flesh, peel and seeds extracts of pumpkin by <br />
<br />
CUPRAC method. DPPH and CUPRAC methods gave linear result in antioxidant activity of flesh and <br />
<br />
peel extracts of pumpkin. <br />
