Description
Summary:Free radical is compound which has unpaired electron on their outer orbital, so it became reactive.

An antioxidant is a compound that donates electrons so it can inactivate the development of

oxidation reactions by preventing the formation of free radical. A lot of plants having antioxidant

activities, one of them is kepundung (Baccaurea racemosa Müll.Arg.). Leaves of kepundung use to

cure constipation and facilitate woman through the menstruation period traditionally. The

objectives of this research were to identify the chemical compound from the leaves of Baccaurea

racemosa Müll.Arg., study the antioxidant activity of kepundung leaves through the determination

of IC50 scavenging free radicals activity DPPH, to isolate the antioxidant compound, and to

determine the active compound; fenol and flavonoid total content. Crude drugs of kepundung

leaves were extracted by the reflux method using ethanol as the solvent. The value of IC50 of DPPH

scavenging activity, total phenolic content, and total flavonoid content were 1,86 ± 0,18 µg/mL;

2,36 ± 0,15 g GAE/100 g; 1,93 ± 0,02 g QE/100 g. Extracts were continued to fractionation by liquidliquid extraction using n-hexane and ethyl acetate as solvents. Ethyl acetate fraction was

subfractionated using radial chromatography. Subfraction 12 until 50 were purified using TLC

preparative. The purity was observed using three different mobile phases of TLC. The isolate was

characterized using specific spot reagent, ammonia vapour, and spectrophotodensitometry. The

isolate was predicted to be an isoflavone lacking a free 5-hydroxyl group with maximum

absorbances at 263 nm and 328 nm.
