Thrombosis is the formation of blood clots in vessels that involve the process of hemostasis.

Almost all cardiovascular diseases have a correlation with thrombosis. Traditionally binahong,

kemuning, and sambiloto have blood cleansing functions. This study was conducted to determine

the antithrombotic activity of binahong, kemuning, and sambiloto extracts in vivo. Extracts were

obtained by maceration method using 96% ethanol. Antithrombotic activity was carried out by

measuring bleeding time, coagulation time, total bleeding, and bleeding pattern using male Swiss-

Webster mice. In the bleeding time and coagulation time test, the mice were divided into nine

groups, namely the normal group, the aspirin group, the warfarin group, and the extract group

with doses of 140 mg/kg and 280 mg/kg, respectively. Time of bleeding, coagulation time, total

bleeding, and bleeding pattern was measured 30 minutes after administration of the test

substance on day 3. From the results of the bleeding time and coagulation time, kemuning extract

140 mg/kg and 280 mg/kg significantly extended both parameters and sambiloto extract 280

mg/kg extended the bleeding time. When bleeding and normal group coagulation time, kemuning

extract 140 mg/kg and 280 mg/kg respectively were 55.33 ± 2.52 and 56.83 ± 4.65 seconds, 86 ±

12.16 and 77.33 ± 19, 53 seconds, 167 ± 40.04 and 89.67 ± 5.62 seconds, and bleeding time of

sambiloto extract 280 mg/kg is 86.67 ± 19.86 seconds. From the results of the total bleeding test,

binahong extract 280 mg/kg and kemuning extract 280 mg/kg increased the total hemoglobin

level with a value of 14.21 ± 3.53 g/dl and 27.92 ± 4.99 g/dl compared to the normal group with a

total hemoglobin level of 0.91 ± 0.41 g/dl. The bleeding pattern shows rebleeding of binahong

extract 280 mg/kg group in the 15th minute and kemuning extract 280 mg/kg in the 25th minute.

Kemuning extract 280 mg/kg gave the best antithrombotic activity in all parameters tested.

Binahong extract 280 mg/kg tends to be antithrombotic in total bleeding test. Sambiloto extract

280 mg/kg antithrombotic activity only observed in bleeding time.
