It is highly contagious and dangerous to young children. Pertussis can be prevented by immunization with pertussis vaccine. The bacteria culture composing pertussis vaccines is obtained by cultivating B. pertussis, which is known to be slow growth bacterium, therefore the yield is usually low. The objective of this research is to increase the biomass from B. pertussis cultivation. It was observed that increasing the growth medium to 200% dan 300% did not improve the biomass of the batch culture. Batch cultivation in bioreactor resulted biomass of 0.48 g/L at 60 hours of cultivation. Applying fed-batch cultivation with growth medium as feed medium at the flow rate of 200 mL/hr also gave 0.48 g/L of biomass at 60 hours of cultivation. Fed-batch cultivation of feed rate 200 mL/hr and oxigenation slightly increase biomass to 0.53 g/L at 60 hours of cultivation. Decreased feed rate of 45 mL/hour resulted in biomass of 0.54 g/L at 60 hours of cultivation. Decreased feed rate of 45 mL/hour together with lowering the pH of feed medium to 5.10 increased the biomass to 0.69 g/L after 60 hours of cultivation. Adjusting culture pH manually to 7.0 – 7.5 by addition of HCl 1 M at sampling intervals resulted in biomass of 0.68 g/L at shorter duration of cultivation, i.e. 48 hours. text
